研究领域
细胞器选择性自噬与疾病发生:选择性自噬通过受体分子将特定的蛋白或细胞器靶向到自噬小体中进行降解,以维持细胞稳态。它与多种疾病(如代谢类疾病、癌症、神经退行性疾病)的发生关系密切。内质网作为细胞内的重要细胞器,其选择性自噬降解机制受到了高度关注,但目前内质网自噬的调控机制及生理病理功能还很不清楚。我们前期利用酿酒酵母进行大规模遗传筛选鉴定了多个全新的内质网自噬因子,如Lst1/SEC24C-Sec23促进内质网片段化来促进其自噬性降解,并可促进引起遗传性肝病的甲抗胰蛋白酶Z突变体降解;膜转运蛋白UVRAG促进内质网自噬起始点组装,并可促进引起早发型糖尿病的胰岛素突变型前体Akita的降解。实验室综合运用酵母、细胞、果蝇和小鼠为模型来研究细胞器自噬(如内质网自噬)的分子机制,及其与疾病发生的关系。
